The corpus spongiosum, the part of the penis that makes up most of the shaft and provides erectile function, is a porous tissue. By design, the overall tissue density increases when the spongiosum fills with blood during sexual arousal (or artificially by a saline solution, for example). As more blood enters the tissue, the penis become erect and hardens. So yeah, it could refer specifically to the mass per unit volume of the person's penis. But more likely, they are using "massive" in more of a metaphorical way, to imply that the penis is capable of seriously hurting the women it's about to rape.
